The museum is housed in the former Sint Jacobs Gasthuis, a charitable institution originally founded between 1262 and 1272. The current structure dates from a reconstruction between 1786 and 1789 in the Neoclassical style, following the demolition of the original medieval building. A major architectural transformation occurred in 2014, designed by MVRDV, which converted the central neoclassical Giudici chapel into a new entrance, shop, and café area. This intervention introduced a distinct red shelf element that houses new functions while respecting the historic monument's austere character.

The museum's collection is divided into two main pillars: modern and contemporary art, and cultural history. The art collection is renowned for its strong holdings in the Cobra movement, featuring works by artists such as Karel Appel, Constant, and Corneille, alongside significant pieces by Lucebert. The collection strategy historically prioritized acquiring works from active movements, including informal art, abstract expressionism, and systematic geometric abstraction from the 1960s. The historical collection comprises over 1,000 objects detailing the city's past, including specific utensils and artifacts related to local industry and daily life.
