The art collection was inaugurated in 1913, coinciding with the completion of the Palace of Culture. The initial Pinacoteca collection comprised 68 works received in custody from the National Museum of Art in Budapest. These were augmented by 15 additional paintings purchased by the art-loving mayor György Bernády using funds from the free royal city of Târgu Mureș. A significant period of growth occurred between 1932 and 1940 under the custodianship of artist Aurel Ciupe, who facilitated the acquisition and donation of Romanian art. Following World War II, the collection expanded further through donations from artists like Ion Vlasiu and collectors such as the Baciu and David families.

The museum is situated within the Palace of Culture, a representative work of the Art Nouveau style with European value. Designed by Budapest architects Marcell Komor and Dezső Jakab between 1908 and 1913, the building features an iconic iconographic program and interior decoration by members of the Gödöllő Artist Colony. Notable architectural and artistic features include the Mirror Hall on the first floor, twelve supreme-value stained glass windows created by Miksa Róth's workshop in 1913, and a monumental Concert Hall with exceptional acoustics housing a Rieger organ inaugurated in 1913.
